Evaluation of 5-[(Z)-(4-nitrobenzylidene)]-2-(thiazol-2-ylimino)-4-thiazolidinone (Les-6222) as Potential Anticonvulsant Agent

It was determined that the studied 5-[(Z)-(4-nitrobenzylidene)]-2-(thiazol-2-ylimino)-4-thiazolidinone (Les-6222) affects cyclooxygenase pathway of arachidonic acid cascade, markers damage to neurons on models PTZ kindling. In model chronic epileptogenesis in mice (pentylenetetrazole kindling), a 4-thiazolidinone derivative showed high anticonvulsant activity, which is weaker than effect sodium valproate and higher Celecoxib. The mentioned compound has pronounced anti-inflammatory brain background kindling, reliably inhibiting COX-1 COX-2. predominant inhibition COX-2 by 44.5% indicates this enzyme’s selectivity Les-6222. According molecular docking study results, revealed properties COX-1/COX-2 inhibitor especially 5-LOX/FLAP. decreasing content 8-isoprostane Les-6222 group beneficial cell membranes oxidative stress during long-term administration PTZ. addition, significantly decreased neuron-specific enolase, indicating neuroprotective epileptogenesis. obtained results experimentally substantiate feasibility further developing as promising agent.
